The idea for the plot should be first. Even if it's a really vague plot, or just a couple of specific details. Then setting. Then characters. For characters, start with what you want the character's personality to be, then design them around that. Then you come up with the entire story, and how the characters will interact. And then you outline the whole thing. Then you start drawing.
