Food, probably. They've already got a ready supply of water thanks to the water purifiers and rainwater catchment built into the school.

If Megu-Nee had just taken everyone to the basement with her, she would be alive right now.
She was this close to surviving too as indicated by that bloody handprint on the medical suitcase containing the anti-zombie medicine before finally succumbing to her injuries and reanimating as a zombie.

Uh, I doubt that. If anything, they'd all be dead. When she locked them in the room and made a run for it, there were still hella zombies wandering around outside.

Megu-Nee was strong. She managed to solo her way to the basement despite being in the school's upper floor where she left her imouto and the other girls.
If Shovel Chick and Closed Eyes were with her with a baseball bat or stick; even if they get bitten, everyone would have a chance to use anti-zombie medicine on themselves in time.
Megu actually receiving backup would have helped her survive instead of being a one-man army that eventually resulted in her being too injured to take the medicine and finally dying down in the basement.
Judging from the flash-back, the school was not as cleared as it is right now.
It would have been a disaster to try and lead the small group to that shelter entry on ground level through the "crowded" corridors
Once you're bitten, it's pretty easy to go full #YOLO and run through a horde of zombies but normally you'd never dare to even try it so it wasn't exactly a feat of her to reach the shelter, besides trying to lead an entire group of healthy persons through the crowded corridors is a recipe for disaster anyway.
Not to mention if Megu-nee just probably never visited the basement shelter and didn't know for sure she'd find potent medicine here, it'd have been one hell of a gamble
When is Rii-san gonna get any development? Out of the four of them, she's the most nondescript (no flashbacks, etc).

The zombies in this story are slow as fuck and there are already chapters showing the girls easily pushing a zombie down even without weapons.
Crowded corridors aren't actually a big deal.
Just makes it easier to trip all the zombies in one go since they are always stumbling around.
Megu-nee read the manual, though, once the zombie infestation started and the manual pretty much tells you that there is anti-zombie medicine in the basement.
The reason she died is because of her thinking that only adults should handle everything and take all the burden upon themselves.
