Homura, for all her issues, is not a killer. The only person she was ever able to kill was Madoka, and that's haunted her since.
There's a difference between being pragmatic and being an unabashed murderer. The only surefire way to kill Mami is to attack her when she's completely unaware, like an assassin. There's no way Homura has that in her. Also, killing Mami wouldn't really serve much of a purpose, since QB would hound Madoka for a contract anyway, and then Kyouko would show up, making Sayaka's issues worse, which only makes Madoka more likely to contract.
Poor Homura, she's the one who tries to reason with Mami countless times, and Mami's response is always hostile.

I think killing Sayaka is entirely in line with her character at that point. Recall that she already mercy-killed Madoka to prevent her from becoming a witch, and she didn't even really have to do that, she could have just run away to the next timeline. Killing Sayaka would essentially also be the same thing, since she was beyond saving otherwise, even if she didn't appreciate it.

Well that's really an argument for not bothering to chase him down in the first place.
Bullets "bouncing around" in the chamber only happens if the bore is larger than the bullet which was required in early smoothbore muskets because they were muzzle loaded and thus needed to have enough extra space to push the bullet to the back, the breech. This problem was solved with rifling and breech reloading and allowed bullets to be larger than the bore diameter. When the propellant was ignited the explosion would heat up the bullet and "melt" it, pushing squeezing it through the smaller barrel's opening where it conforms to the shape of the inner barrel walls and gives it the spin. Now Mami's rifle is clearly muzzle loaded but since she spawns the ball inside I'm not going to question too hard how that works, maybe the breech is slightly larger.
Round bullets require a lower twist rate to stabilize because it's centre of gravity is in the exact middle instead of at the back like modern bullets are with their aerodynamic "sharp" tips (There is a reason arrows are front heavy instead of back heavy, the back will have more momentum thus try to move to the front and unstabilize it). Higher twist rate means lower velocity since it has more friction, so that's bad if you want more range/accuracy, fortunately Mami shouldn't need much. Finally, longer barrels increase velocity which in practice increases accuracy since more energy means less interference from the environment.
Flintlock is stupid though, at least use a percussion cap if you want to be an antique hipster.

Maybe she wanted Sayaka to take her seriously, and the only way for Sayaka to actually believe Homura might be doing this for something good(Madoka) is that she back her logic up with a real intent to kill.
Once she does that, Sayaka has to consider that there is something more to Homura(and then Kyouko) than she thought, instead of assuming they just want grief seeds, power, kill humans to benefit themselves etc.
Maybe you could argue that this action is partially responsible for Sayaka really opening up at the end to Kyouko.
It's not necessarily about faking or bluffing, just that she might have expected Kyouko to jump in. If she really only wanted to kill Sayaka, then she would have dropped that nade earlier and went to shoot her in a time stop.

I think the intention of that scene is to show that murder does not go unanswered. Karma is coming back to hit Sayaka before she kills again.
Many just assumed she didn't kill on the train, which I think was the reason they removed the Homura scene from the movies. The intention was to show Homura killing a witch in the making, something that has begun cursing the world by killing, someone that can no longer be saved.
But some people assume it's Homura acting crazy and killing an innocent, which doesn't seem to be their intention. In the movies, they made Sayaka even more crazy sounding during her fight with Elsa Maria. They even replaced the sad song with one called "She is a witch!" to get the point across. Kept the train scene, removed the Homura threatening Sayaka scene.
