Hey! /g/ here.
I'd like to programmatically control the generation of human models from a set of parameters. For example one parameter controls the color of the shirt and I can loop through a bunch of possible colors in a script and generate images of the model with different color shirts. Same for rotation, joint angles, etc..

In some computer science fields, using SMPL is common (http://smpl.is.tue.mpg.de/), but I still don't understand how to use this in a full pipeline - any pointers, or even words to google would be helpful.

Isn't this possible already with DAZ?
I don't like the software but it is the closest thing to what you are trying to do.
I suppose you want to make a plugin, so, maybe check for automatic rigging plugins for Maya or Blender and see how it is managed
