Is it possible to make a 3d render look like a oil painting just by using the render engine?
Or will it just not look right without manual touch up in a 2d painting software?

Maybe, but a lot of your work would go into how your camera is interpreting the scene. I would say it's would be easier to just render out your separate layers and then composite them in Photoshop or AE, but there's some people out there that prefer the final render to be the final result and will tweak their work flow around that concept. Working in game engines would be a good example of this.
