How do I achieve this look?

I know toon shading is a thing, but seeing a few examples like this take it to a whole different level

look up any toon shader on blender tutorial.

That's why for good looking faux-2D via 3D, you're going to have to hand tweak every frame
Which is a lot of work, but still 1/1000th the work compared to drawing every 2D frame

Posting my own stuff.

Didn't model anything myself, all preexisting stuff compiled in poser and rendered with a toon shader. Lines were created with comic book preview mode with some touching up in SAI. Mouth, nose and winking eye were indeed drawn on but my aim is to make art work that looks as convincingly 2D as possible.

Then forget the shaders, the rendering, and even the scene building; just pose, take a screenshot, and then trace it in The GIMP and dodge/burn to perfection.
